Proliferative Effect of Serum on Bovine Retinal Pigment Epithelial Cell Culture

As serum in the vitreous cavity stimulate the migration and proliferation of RPE cell, serum is necessary to grou RPE cells in culture.
Bovine RPE cells were cultured with various concentration of fetal bovine serum(FBS 0%, 5%, 10%, 20%, 4)%) to evaluate their effects on RPE cell growth.
The mean of second passage RPE cells was called "initial mean cell count".
The all results of experiments were compared with initial mean cell count.
The final cell number was deterined after 7 days using a Coulter conuter.
@EN 1. The FBS stimulated the proliferation of bovine RPE cells, as incresing its concentration except 0% and 40%.
2. The percent proliferation of RPE cells were decreased to 72.6% with 0% FBS and 10.8% with 40% FBS. The percent proliferation of RPE cells were increased to 7.5% with 5% FBS, 68.7% with 10% FBS and 157% with 20% FBS.
It was significantly increased only with 20% FBS(P<0.05).
These results suggest that FBS stimulated the proliferation of bovine RPE cells in a dose-dependent manner, with maximal proliferation at 20% FBS. But at the highest concentration (40% FBS). This proliferative effect was suppressed.
